I am working in Dr. Blanca Barquera's lab where I have been growing and harvesting Vibrio cholerae cells (causes cholera in humans) and then purifying for a membrane bound protein from these cells using chromatography columns. This protein is involved with the sodium ion channels which are correlated with the toxicity of cholera. I have been experimenting with different growing conditions to get the most cells to grow. Some examples are adding arabinose or ethanol at certain times in the growing stage. I have also been trying different purifying conditions (different buffers, amount of resin in the column) to purify the protein. After the protein is purified, I run concentration and enzyme activity assays.
