The optimum component concentrations were found to be:
1.6mg of luciferase/oxidoreductase preparation and 50
mM FMN in the potassium phosphate buffer (100mM; pH 7.8) containing decanal (1mL/L; 6mM) in a 1.0mL total volume. The morphine assay required the addition of 750mM NADP+ and 0.5 unit of morphine dehydrogenase to the optimized luminescence assay in a total volume of 1.0mL.
