Pilot Laboratory Testing Program
The overall objective of the pilot laboratory testing program is to improve the reliability of laboratory results. Currently it is not possible to purchase reference Dreissena larvae samples, so laboratories that are in the process of developing and optimizing existing and new techniques are unable to independently assess their results.
A primary purpose of this pilot testing program is to prepare and make such reference materials widely available.
Dreissena larvae (both D. bugensis and D. polymorpha) of various sizes will be collected and manually isolated. At the request of research and analytical laboratories, agencies contracting analytical services, and monitoring programs we will provide veliger samples as either spiked standard reference samples (with known #’s of veligers) or spiked blind samples (with numbers of veligers not provided until after samples have been analyzed). Veligers will be spiked into a sample matrix (i.e. plankton sample) either provided by the supplier or provided by the requesting laboratory. Upon completion of sample analysis, and reporting of results, individual labs will be provided a report on the accuracy of their analyses. Also, all data generated and reported will be entered into a central database (with lab identities kept confidential) to assist in a larger goal of identifying optimal analytical methods for the early detection of Dreissena larvae in plankton samples.
Preparation and Distribution of Reference Materials:
Dreissena larvae from Western waters including Lake Mead, NV and Lake Mohave, AZ where D. bugensis are abundant and D. polymorpha is absent.
Dreissena larvae from Lake Champlain, VT and Lake Saratoga, NY where D. polymorpha larvae are abundant and D. bugensis is absent.
Larvae from these samples have been archived in 25% and 70% ethanol.
Upon request --Standard reference samples spiked with larvae in low (0-5 l-1) or high (15-25 l-1) will be prepared in either plankton material provided by the participating laboratories or plankton material we have obtained from (xxx, GA) “certified” to be Dreissena-free prior to spiking.
Upon request-- Blind samples spiked with larvae in low (0-5 l-1) or high 15-25 l-1 concentrations will be prepared in either plankton material provided by the participating laboratories or plankton material we have obtained from (xxx, GA) “certified” to be Dreissena-free prior to spiking.
